摘要
目的 :研究持续恒压组织扩张术 (CPTE)和常规间断组织扩张术 (CITE)对成纤维细胞胶原合成能力的影响。 方法 :用 12只白色小家猪制作 CPTE和 CITE动物模型 ,用核酸分子原位杂交和免疫组织化学技术观察成纤维细胞前胶原 m RNA和前胶原蛋白的表达。 结果 :扩张中成纤维细胞内前胶原 m RNA和前胶原蛋白的表达增强 ,扩张皮瓣移植后进一步增强 ,皮肤厚度恢复后下降 ,CPTE组的表达强度高于 CITE组。皮瓣形成后 CTPE组的真皮厚度恢复时间早于 CITE组。 结论 :终止扩张后成纤维细胞前胶原 m RNA和前胶原蛋白的表达高于扩张期。
Objective:To investigate Effects of continuous pressure-controlling tissue expansion(CPTE) and conventional intermittent tissue expansion(CITE)on expression of procollagenⅠ,Ⅲand mRNA in fibroblasts. Methods:Twelve pigs were chosen for CPTE and CITE animal model.Expanded skin was examined with nucleic acid hybridization and immunocytochemistry techniques. Results:The expression of procollagen and mRNA enhanced significantly during expansion,increased more after expanded flap insertion.and decreased after thd skin thickness recovered.The expression of procollagen and mRNA in CPTE group was higher than in CITE group.The decreased skin thickness in CPTE group recovered earlier than in CITE group. Conclusion:The expression of procollagen and mRNA is enhanced during and after expansion.Continuous pressue-controlling tissue expansion reduce tissue growth more effectively.
