电针及生物活性物质体外孵育对大鼠淋巴细胞中介免疫的影响

EFFECTS OF ELECTROACUPUNCTURE AND INCUBATION IN VITRO WITH BIOACTIVE SUBSTANCES ON CELL-MEDIATED IMMUNITY IN THE RAT LYMPHOCYTE

摘要 用酸性非特异性酯酶组化染色法(ANAE)观察了电针刺激(Ea)及经外源性5-羟色胺(5-HT)、P物质(SP)、β-内啡肽(βEP)和甲硫脑啡肽(MEK)孵育后的电针组、纳洛酮阻断电针组及对照组的淋巴细胞分型的改变,以及大鼠淋巴细胞与人食管癌上皮细胞(Eca-109)共育前后ANAE分型的变化。结果表明:①电针可使大鼠淋巴细胞ANAE点样型(ANAE—F^+)增多(P<0.05),对ANAE弥散型(ANAE—D^+)影响不显著(P>0.05)。几种活性物质对ANAE—F^+和ANAE—D^+的影响与电针效应一致,纳洛酮可阻断此作用。②电针、纳洛酮阻断电针及对照组三组淋巴细胞(无论经活性物质孵育与否)与Eca—109细胞共育后,ANAE—F^+减少(P<0.0005),ANAE—D^+增多(P<0.0005),部分ANAE—D^+细胞有形态学改变。由比提示,电针及活性物质孵育对淋巴细胞ANAE分型的影响可能是通过内源性阿片样肽(OLP)受体起作用,肿瘤细胞与淋巴细胞共育可显著影响淋巴细胞的功能活性。 We studied the effects of Electroacupunture (Ea) and of β-endophin (βEP),Met-enkephalin (MEK), Substance P(SP) and 5-Hydroxytryptamine (5-HT) on the cell mediated immunity in rat lymphocytes, with the histochemical staining of a-naphthyl acetate esterase (ANAE). The changes of the ANAE patterns in the Ea, Naloxone (NX) and control groups before and after co-incubation with Eca-109 cells were detected as well. The results showed ①Ea could enhance the ANAE focal pattern(ANAE-F+) (P<0.05) but had no significant effect on the ANAE diffuse pattern(ANAE-D+).NX could inhibit the effect. BEP, MEK, SP and 5-HT caused the same changes in both ANAE-F+ and ANAE-D+ as Ea did. ②The ANAE-F+ was lower and ANAE-D+was higher after co-incubation than before. Some of the ANAE-D+ pattern ymphocytes showed changes in morphology. The results suggest that the effects of Ea and of incu bation with BEP, MEK, SP and 5-HT on the ANAE pattern may be mediated by opioid like peptide (OLP) receptor, and co-incubation with Eca-109 cells may change the lymphocytes' functional activities.
