摘要
目的 :为观察中药平颤汤治疗帕金森病的药理作用 ,探索其作用机理 ,本文主要观察平颤汤对自由基的影响。方法 :从大鼠肝脏、大脑分离线粒体 ,用Fe2 SO4-VitC处理 ,分别加入平颤汤或其 3种拆方 ,测定MDA的变化 ;给大鼠平颤汤灌胃 4d ,用 6 -OHDA立体定向损毁左侧黑质 ,继续给药 4d后处死 ,取黑质和纹状体 ,测定MDA、T -SOD、GSH、GSH -PX ;用 6 -OHDA损毁大鼠黑质造成自由基损伤模型 ,给平颤汤灌胃 10d后处死 ,测定黑质和纹状体中MDA、T -SOD、GSH、GSH -PX。结果 :离体实验预先加入平颤汤可明显抑制FeSO4-VitC引起的肝脏、大脑线粒体脂质过氧化 ,使MDA减少 (P <0 0 0 1) ;在体实验预先灌胃平颤汤 ,再遭受 6 -OHDA的氧化损伤鼠 ,黑质和纹状体中GSH和GSH -PX明显增高 (P <0 0 1) ,MDA降低 (P <0 0 1) ;先用 6 -OHDA造成黑质自由基损伤 ,再灌胃平颤汤的大鼠 ,其黑质和纹状体中MDA明显下降 (P <0 0 0 1) ,GSH和GSH -PX明显上升 (P <0 0 0 1)。结论 :平颤汤具有抗自由基氧化损害 。
Objective:To observe the pharmacological effect of Pingchan decoction on the treatment of Parkinson disease and research its mechanism,this paper mainly studied the influence of Pingchant decoction on free radical.Methods:Separating the mitochondria from the liver and brain of the rat,processed by Fe 2SO 4-VitC,adding Pingchan decoction and its three similar prescriptions,the changes of MDA were measured.The rat was filled stomach with Pingchan decoction for four days.After the left black substance was destroyed by stereotaxic technigue with 6-OHDA,the rat was given continually medicine for four days,then killed,taking out the black substance and striate body;MDA,T-SOD,GSH,GSH-PX were measured.The black substance was destroyed by stereotaxic technique with 6-OHDA and the injured model by free radical was made,the rat was given filling the stomach with Pingchan decoction for ten days,then killed,and MDA,T-SOD,GSH,GSH-PX in the black substance and striate body were measured. Results:After taking Pingchan decoction the isolated experiment can obvilusly inhibit the peroxidation of mitochondrial lipid of the liver and brain caused by Fe 2SO 4-VitC,reducing MDA(P<0 001) The experimental rat in vivo was filled the stomach,then was injured by the oxidation of the 6-OHDA,the GSH and GSH-PX in its black substance and striate body were apparently increased(P<0 01) and MDA decreased (P<0 01) After the black substance was injuried by free radical with 6-OHDA,the rat was given filling the stomach with Pingchan decoction,and MDA in its black substance and striate body was obviously decreased (P<0 001);GSH and GSH-PX were apparently increased(P<0 01) Conclusion Pingchan decoction plays role in antioxidation injury of free radical and in protection the neuron.
