摘要
天花粉蛋白是从中草药禾古楼根中提取的一种核糖体失活蛋白 ,具有致流产、抗肿瘤和抗 HIV功能。本文应用共聚焦激光扫描显微术结合特异性荧光探针 2′,7′-二氯荧光黄双乙酸酯和 Fluo3- AM,观察了天花粉蛋白作用于小鼠胚胎细胞过程中活性氧自由基和细胞内钙离子浓度的变化。实验结果表明 ,天花粉蛋白能诱导小鼠胚胎细胞钙浓度升高和产生活性氧自由基 ,并且天花粉蛋白能有效抑制小鼠胚胎细胞的分裂 。
Trichosanthin, a ribosome inactivating protein extracted from the root tuber of a traditional Chinese medicinal herb Trichosanthes kirilowii, possesses abortifacient, anti tumor and anti human immunodeficiency virus (HIV) activities. With confocal laser scanning microscopy in combination with the fluorescent probes 2′, 7′ dichlorofluorescin diacetate (H 2DCF DA) and Fluo 3 AM, trichosanthin induced changes in reactive oxygen species and intracellular calcium in mouse embryos were observed. The results indicated that trichosanthin induced increase in intracellular calcium and production of reactive oxygen species in mouse embryos, and that trichosanthin effectively inhibited the development of mouse embryos. This provides new insight into the mechanism for abortifacient activity of trichosanthin.
