摘要
目的:探讨分离外周血细胞的方法及该方法在免疫偶合物制备过程中的应用价值。方法:应用 Ficoll/Hypaque分离液经密度梯度离心得到单个核细胞、红细胞及混有红细胞的粒细胞;用 0. 83%氯化铵溶解混杂的红细胞得到粒细胞;利用单核细胞粘附玻璃的特性除去淋巴细胞中的单核细胞;用 AET-E玫瑰花试验分离 T淋巴细胞及 B淋巴细胞。将分到的各血细胞与单克隆抗体及免疫偶合物反应,活细胞免疫荧光法测定。结果:分到的各种血细胞 2h活性达 96%以上,细胞得量较高。所测定的单抗特异性高,活性保持良好。结论:该法是一种简单易行、经济方便的方法,可用于免疫偶合物的制备研究中。
: objective:To study the method of isolating the cells from the peripheral blood and the application in the study of immunoconjugate.Methods:The mononuclear cells,erythrocyte,granulocyte were gained by density gradient centrifugation using Ficoll/Hypaque.Erythrocytes among granulocytes were destroyed by 0.83% of NH4Cl.And monocytes mixed with lymphocytes were removed by adhering glass of monocytes. T and B lymphocytes were separated by using the AET-E rose forming test. Results:Various blood cells from the peripheral blood kept over 96% of the activities. Great quantities of the cells were gotten.The monoclonal antibodies and the monoclonal antibodies in the immunoconugates kept their good activities specificities. Conclusions:It is a easy,simple,economic and convenient method.It can be used in the study of preparing immunoconjugates.
