IFN-γ作用于沙眼衣原体感染细胞后Fas mRNA的检测 被引量:1

EXAMINATION OF FAS mRNA AFTER ADDING GAMMA INTERFERON TO Chlamydia trachomatis-INFECTED CELLS

摘要 探讨了IFN γ能否通过上调Ct(Chlamydiatrachomatis ,简称Ct)感染细胞FasmRNA的表达而调节机体对Ct感染细胞的免疫应答。应用IFN γ作用于感染Ct (K血清型 )的McCoy细胞 ,通过观察细胞形态改变及MTT法检测细胞毒作用 ,选出IFN γ的最佳作用浓度和检测时间 ,用RT PCR方法检测McCoy细胞FasmRNA的表达。结果显示 :(1)形态学观察结果显示IFN γ作用于Ct感染细胞 2 8h后细胞膜下出现大量空泡 ,整个细胞内充满颗粒 ,且随着IFN γ作用时间延长 ,细胞体积变小 ,细胞变稀疏。 (2 )MTT法检测结果显示 2 0U/mLIFN γ对Ct感染的McCoy细胞已产生显著的细胞毒作用 ,IFN γ浓度继续增大 ,细胞毒作用的增大不明显。 (3)RT PCR法检测FasmRNA的表达 ,结果显示IFN -γ上调了McCoy细胞FasmRNA的表达 ,Ct感染McCoy细胞没有影响FasmR NA表达的上调。IFN Contribution of Gamma interferon to control immune responses to Chlamydia trachomatis infected cells by up regulation the expression of Fas mRNA was studied. The optimum concentration of IFN γ and exposure time were screened to the C. trachomatis (serotype K) infected McCoy cells and treated with IFN γ through observing the morphological change of cell and detecting the IFN γ mediated cytotoxicity to the infected cells by MTT assay, and the expression of Fas mRNA on McCoy cells analyzed by RT PCR. A large number of bleb appeared at 28 hour under the infected cells' surface after treatment with IFN γ, and particulate matter spreading the whole cells was observed in the infected McCoy cells. So long as the prolonging exposure time, the cells lost their volume, and became scattered. 20 U/mL of IFN γ was the optimum concentration; and as the IFN γ concentration increased the cytotoxicity did not increase remarkably. RT PCR analysis revealed that IFN γ could increase the expression of Fas mRNA in the infected cells. And the infection of C. t. did not affect the up regulation of the expression of Fas mRNA. Therefore, IFN γ might up regulate the expression of Fas mRNA to control immune responses to the C. t. infected McCoy cells.
