摘要
目的研究 C2基因在胃癌中的表达、分布及意义.方法用免疫组化(ABC 法)方法检测 C2在58例胃癌(男45例,女13例;年龄≤50岁19例,>50岁39例;高分化14例,中分化21例,低分化23例)及其44例癌旁组织中蛋白水平的表达,RT-PCR 法检测 C2mRNA 在人胃癌细胞系(7901,VCR,MKN,803)中的表达.结果 C2蛋白阳性率为41.4%(24/58),44例其癌旁组织中,C2蛋白阳性率为77.3%(34/44).χ~2检验:C2蛋白在胃癌组织中表达明显低于其癌旁组织(X^2=11.718,P<0.001),且与胃癌组织的分化程度、浆膜浸润、肿瘤大小有明显的相关性[X^2值依次为18.977(P<0.001)、14.294(P<0.001)、8.108(P<0.004)]RT-PCR 结果显示在四种人胃癌细胞系中均可检测到 C2mRNA.结论 C2基因表达下调可能与胃癌的发生。
AIM To study the distribution,expression and significance of translation initiation factor C2 gene in gastric cancer tissue. METHODS ABC immunohistochemical method was used to detect the expression of C2 protein in 58 samples of gastric cancer tissue (males:45 samples,females:13 samples;age≤50 years:19 samples,>50 years:39 samples;high grade:14 samples,middle grade:21 samples,low grade:23 samples)and 44 samples of tumor- surrounding tissue,and RT-PCR method was used to detect the expression of C2 mRNA in human gastric cancer cell lines(7901,VCR,MKN,803). RESULTS In 58 samples of gastric cancer tissue and its 44 samples of tumor-surrounding tissue,the positive rates of C2 protein were 41.4% and 77.3%,respectively. Statistical analysis suggested that the positive rates of C2 protein were all significantly higher in tumor-surrounding tissue than that in gastric cancer tissue(X^2=11.718,P<0. 001).C2 protein was significantly correlated with different pathological grading,tumor size,the depth of invasion[X^2=18.977(P<0.001),14.294(P<0.001)、8. 108(P<0.004),respectively].The result of RT-PCR showed that expression of C2 mRNA could be detected in all four human gastric cancer cell lines. CONCLUSIONS Low expression of C2 gene might play an important role in the development of gastric cancer.
