一氧化氮合酶抑制剂对培养的人分泌中期子宫内膜细胞内钙的影响

摘要 目的 观察已受雌、孕激素影响的人子宫内膜细胞在一氧化氮(NO)作用下的细胞生理变化。方法 取人分泌中期子宫内膜,体外原代培养24-48h,用钙荧光探剂Fluo-3-AM负荷钙离子,采用激光扫描共聚焦显微镜测定加入一氧化氮合酶抑制剂(L-NAME)后,子宫内膜细胞内游离钙离子浓度([Ca^2+]i)的动态变化。结果 加入L-NAME后,腺上皮细胞和间质细胞[(Ca^2+]i立即开始升高,400s后上升至高峰,900s时稍有下降,然后继续升高并保持在高水平,同时发现腺上皮细胞和基质细胞对L-NAME的反应不同。结论 在雌、孕激素作用的基础上,NO通过改变子宫内膜细胞[Ca^2+]i进行信号转导,实现其生理效应。  Objective To investigate the effect of nitric oxide (NO) synthase inhibitor (L-NAME) on the intracellular free calcium concentration of cultured human endometrial cells. Methods Human endometrial cells were obtained from patients in the implantation window phase of their cycle during endometrium biopsy and were cultured in vitro. After 24 to 48 h of culture, the cells were incubated with Fluo-3-AM, and then stimulated by L-NAME. The dynamic change of intracellular calcium concentration ([Ca2+]) were determined by laser scanning confocal microscopy. Results After the addition of L-NAME, [Ca2+]i was immediately increased, and reached its peak value at 400 s. At 900 s [Ca2+]i was slightly decreased, then rose again and maintained at a high level within the testing period. In addition, it was observed that the increasing range of [Ca2+]i in epithelial glandular cells was significantly higher than that of stromal cells. Conclusion L-NAME could increase [Ca2+]i in endometrial cells , which would probably involve the mechanism of the signal transduction in the endometrium.
