摘要
目的 研究HHK-Ⅱ人工腱植入后不同时期泛素(ubiquitin)的表达与分布。方法 将兔后腿屈肌腱切除一定长度后用HHK-Ⅱ人工腱连接,不同时间后用免疫组织化学法检测泛素在人工腱及其周围组织中的表达与分布,并与正常肌腱组织对照。结果 在正常自体腱组织中,泛素低表达,且均匀分布;HHK-Ⅱ人工腱植入后3周,人工腱周围及其附近的新生腱组织中泛素表达为强阳性;植入后6周及9周泛素的表达明显下调 (P< 0.05),其中9周的泛素表达接近正常腱组织。即在人工腱降解期泛素表达为强阳性,自体腱形成期及之后的时期泛素的表达明显下调。结论 人工腱角蛋白的降解可能主要由泛素承担;人工腱周围新生组织中不正常蛋白的清除除有溶酶体参与外,泛素也起主要作用。
Objective To study the expression and distribution of ubiquitin at different phases after the implantation of human hair keratin (HHK-II) artificial tendon. Methods The flexor tendon of rabbit posterior limb was partly removed and replaced by HHK-II, and the expression and distribution of ubiquitin in the implanted artificial tendon and the surrounding tissues were determined by immunohistochemical technique and compared with those in the autogenous tendon tissue at different time points after the surgery. Results In the autogenous tendon tissue of the rabbits, only low levels of evenly distributed ubiquitin was observed. Three weeks after the implantation, high expression levels of ubiquitin were detected in the surrounding tissue of the graft and in the newly generated tendon tissues, and a subsequent decrease took place at the sixth week (P<0.05) till a level close to that in the normal tendon tissues was reached at the ninth week. Conclusion The degradation of HHK-II is undertaken mainly by ubiquitin which also shares with lysosome the task of eliminating abnormal proteins in the newly generated tissues around the HHK-II graft.
