摘要
以微孔滤膜为抗原栽体,用酶标记的单克隆抗体直接检测病死雏鸭组织和鸭胚(或鸡胚)尿囊液中的雏鸭肝炎病毒(DHV-1)。建立了抗原斑点试验(AST)。试验的最佳工作条件:以50%脱脂牛奶,37℃1小时封闭;酶标单抗浓度为1:1600;底物缓冲液为0.05mol/LpH7.6Tris-HCl。用该法检测了175份病料,并进行阻断试验、交叉试验、平行试验及重复性试验。结果证明,这是诊断雏鸭肝炎病毒的一种微量、简便、特异性强的方法。
This paper establishes the monoclonal antibody-antigen spot test (McAb-AST) using nitrocellulose membranes as the antigen carrier with an attempt at detecting the duck hepatitis virus type Ⅰ (DHV-Ⅰ) in liver suspension and allantoamniotic fluid. The optimum working factors of the test have been set up. 50% skim milk as blocking solution acting at 37℃ for one hour may get best result; enzyme-monoclonal antibody conjugate may be diluted to 1:1600 in PBS; and the substrate buffer solution may be prepared by 0.05 tool/L pH 7.6 Tris-HCl. 175 specimens have been detected, the result of block test, cross test and repeat test showing that the McAb-AST is simple, trace and specifie for diagnosis of duck viral hepatitis.
