摘要
目的 观察血管紧张素Ⅱ (AngⅡ )对培养牛主动脉血管平滑肌细胞 (VSMC)增殖的作用及银杏内酯对其增殖的影响。方法 采用组织块贴壁法培养牛主动脉VSMC ,以3 H TdR掺入法和流式细胞仪分别测定VSMC的DNA合成量及细胞周期中各期细胞构成比。结果 AngⅡ(0 1μmol/L)作用 2 4h使3 H TdR掺入量比对照组增加 34 % ;S期和G2 /M期细胞增多 ,细胞增殖指数由 35 15 %增至 45 0 3%。与AngⅡ组相比 ,AngⅡ加银杏内酯B(G B ,0 1~ 10 μmol/L)组、AngⅡ加G B和银杏内酯A(G A)的混合物 (G AB ,0 1~ 10 μmol/L)组 的3 H TdR掺入量减少 ;S期细胞构成比和细胞增殖指数减少。结论 AngⅡ对VSMC有促增殖作用 ,可被G B和G
Objective To investigate the effects of ginkgolides on cultured calf vasuclar smooth muscle cells(VSMC) proliferation induced by AngⅡ.Methods Calf aortic VSMCs were cultured.The amount of DNA synthesis and cell cycle distribution was measured by 3H-TdR incorporation and by flow cytometry respectively.Results 24h exposure of VSMC to AngⅡ(0 1μmol/L) caused an increase in 3H-TdR incorporation,cell mitogenic activity also increased.The proliferation was inhibited by G-B(0 1~10μmol/L) and G-AB(0 1~10μmol/L).Pretreatment with G-B and G-AB reduced percentages of S period cells and cellular mitogenic activity induced by AngⅡ.Conclusion G-B and G-AB antagonized the proliferation of VSMC induced by AngⅡ.
