摘要
目的 研究转人 TNFα基因对肾细胞癌 ( Renal Cell Carcinoma,RCC)细胞 ( 786- 0细胞 )癌基因表达的影响。方法 将人 TNFα基因与逆转录病毒载体 p LNCX连接转染包装细胞 ,再感染 RCC细胞 ,MTT法测其中 TNFα的生物学活性 ,用免疫组化法检测细胞癌基因的表达情况。结果 转人 TNFα基因 RCC细胞除 bcl- 2的表达没有影响外 ,EGFR、p1 85 erb B- 2和 c- met的表达有一定程度的降低。结论 该结果为转人
Objective To study the influence on the oncogenes expression of the human renal cell carcinoma (RCC) 786\|0 cells transduced with human TNFα gene.Methods Recombinant retroviral vector pL(TNFα) SN was constructed and transfected into the packaged cell.The confirmed retrovirus were infected to RCC cells,and the biological activity of rhTNFα was tested by MTT method.Then,oncogenes expression and TNFα transduced RCC cells were observed by immunohistochemistry.Results The expression level of bcl 2 was not affected,while the expression of EGFR?p185erbB 2 and c met were decreased.Conclusion This study provides partial theory basis for RCC gene therapy by TNFα.
