单克隆抗体亲和层析法纯化疟原虫表面抗原-环孢蛋白的初步研究 被引量:1

摘要 用MS1-Sepharose4B亲和层析柱能自子孢子可溶性蛋白中提取CSP。纯化CSP的分子量约40kd,具有与全虫相同的抗原性,且能分别与MS1-5中任意一株单抗的两个分子同时结合。表明具有重复性表位的CSP抗原决定簇是约氏疟原虫子孢子表面具有免疫原性的主要结构。 Circumsporozoite protein (CSP) ,with molecule weight of 4oKd, was purified from sporozoite extract through MS1-Sepharose 4B affinity column. This was demonstrated that the immunodominant region of CSP molecules is multivalent with regard to the expression of a single epitope by the ability of monomers of CSP to bind simultaneously two or more molecules of the same monoclonal antibody (MS1-5) . Immu-nogenecity of CSP was same as that of sporozoite. It appears that CSP is the most im-munogenic constituents of sporozoite.
