摘要
本实验应用细胞融合技术,首次在国内建立了5株分泌抗约氏鼠疟(BY265)子孢子单克隆抗体的杂交瘤细胞系MS1-5。鉴定结果表明:(1)MS1-5能使子孢子表面染以三种着色方式的特异性荧光,其抗体活性能被全虫子孢子所阻断。(2)各株单抗之间对抗原能产生竞争性抑制,提示MS1-5是针对子孢子表面同一个抗原决定簇的。(3)免疫印渍结果表明,MS1、3、4、5均识别分子量为40Kd的CSP蛋白,除此之外,MS2、4尚能与72和64Kd的蛋白结合。根据它们同样能与MS1、3、5竞争结合抗原的事实,推断MS1-5能识别存在于不同分子上的共同抗原位点。
Hybrid cells(MS1-5) secreting antibodies against sporozoites of p. yoelii were obtained by fusion of sp2/0 myeloma cells with immune murine spleen cells. McAbs (MS1-5) were eualuated for characterizations ( 1 ) All five were position in the indirect immunfluoresoent antibody test with P. yoelii sporozoite, however, which showed the three different immunofluoscence pattern. The activities of MSl-5 were significantly decreased after absorption by sporozoites. (2) The results of competition binding assays indicated that the single immunodominant regions of CSP were recognized by the homologous McAbs MS1-5.(3)Electrophoretic immunoblotting analysis showed that the MS1. 3. 5 were specific for s-porozoit antigen with polypeptidos of 64Kd or 72Kd.
