摘要
本文报告了癌胚抗原时间分辨荧光免疫测定法。采用McAb包被微量滴定条,以双功能基团螯合剂标记Eu^(3+)为示踪物。本法最小检出值为0.7ng/ml,标准曲线范围为2.5~500ng/ml,批内和批间CV平均为7.09%和10.56%,回收率为106.00%。测定方法简便快速,每天一个技术人员可完成300份样品。
A time-resolved fluoroimmunoassay for human carcinoembryonic antigen was develeped. Two kinds of antibody are used, mouse monoclonal antibodies (C50, C6) for coating microtiter strip wells and polyclonal antibodies for Eu3+ chelate labelling. Eu8+ ions are bound to the antibody molecules by means of the anhydride of diethylene-triaminepentaacet acid. The solid-phase sandwich procedure is performed in two steps. The sensitivity of the assay is 0.7ng/ml, with a working range of 2.5-500ng/ml. The mean CV values of intra assay and inte rassay are 7.09% and 10.56%, respectively. Mean recovery of added CEA is 106.00%.The assay is easy and rapid to perform, and is capable of processing in excess of 300 specimens within a working day.
