摘要
由于具有时间、组织和位点特异性 ,cre重组酶介导的DNA重组已成为基因靶位操作的一个重要工具。概述了cre/loxP系统的作用特点 ,cre重组酶的结构和重组机制 ,表达载体构建及重组个体检测等方面的问题 ,重点介绍了cre/loxP系统在基因重组研究中的应用及近来的发展与成就。
DNA Recombination mediated by cre recombinase has become an important tool for gene targeting because it is time-specific, tissue-specific and site-specific. In this paper, the recombinent characteristics of the cre/loxP system,the structure of cre recombinase and its recombination mechanism, the construction of expression vector and the detection of the result of recombination in cre/loxP system is reviewed.The ultilization of cre/lox P system and its recent development and achivements are also introduced as an important aspect.
