过敏性哮喘大鼠大脑和肺组织c-fos蛋白的表达 被引量:5

Expression of c-fos protein in brain and lung in ovalbumin sensitized rats

摘要 目的 探讨哮喘大鼠肺脏和大脑中c fos表达的分布及其意义。方法 制作卵蛋白致敏大鼠哮喘模型 ,采用免疫组织化学ABC方法和图像分析等技术观察Fos蛋白在肺脏和大脑内的分布情况。结果 哮喘组大鼠 ( 1 0只 )肺脏和大脑中c fos表达与正常对照组大鼠比较 ,差异有显著性(P <0 0 5) ;Fos阳性产物在脑内主要集中分布在额顶皮质、边缘前脑 (扣带皮质、梨状皮质和中央杏仁核等 )、丘脑室旁核、下丘脑室旁核、视上核、下丘脑外侧区、下丘脑室周核、孤束核、最后区和延髓腹外侧区内 ;小脑内无明显Fos分布密集区。结论 哮喘的发病机制中可能存在神经免疫调节的参与 ; Objective c-fos expression and its distribution in brain and lung of ovalbumin sensitized and challenged rats were investigated.Methods Asthma rat model sensitized and challenged by ova lbumin was established. c-fos expression was detected with immunohistochemical ABC and image analysis methods. Results c-fos positive s taining increased significantly in both brain and lung in asthmatic group as com pared with normal control. (P<0.01). c-fos positive products were mainly concentrated in frontal and parietal cortex, forebrain limbic system (ci ngulated cortex, piriform cortex and central amygdaloid nucleus), paraventricula r thalamic nucleus, hypothalamic paraventricular nucleus, supraoptic nucleus, la teral hypothalamic area, periventricular hypothalamic nucleus, nucleus of solita ry tract, area postrema and ventrolateral medulla. No distinct gathering area of c-fos protein was found in the cerebellum.Conclusion It is suggested that neur oimmunomodulation may play a role in the pathogenesis of asthma and c-fos prote in might be involved in the neuroimmunomodulation.
