摘要 目的 :观察蝎毒多肽 (APBMV)对鼻咽癌CNE 2Z细胞株细胞膜电位的影响。方法 :细胞经培养传代 ,分组处理。空白对照组加生理盐水 ,其他 3组分别加入浓度为 8,12和 16mg/L的APBMV。采用玻璃微电极细胞内记录法测定CNE 2Z细胞膜电位 ,MTT法观察APBMV对CNE 2Z细胞的毒性作用。结果 :CNE 2Z细胞经APBMV处理48h后 ,细胞膜负电位 (13 0 0± 3.5 8)mV明显小于空白对照组 (2 3 0 0± 8.2 1)mV ,P <0 .0 1,细胞膜呈去极化状态 ;CNE 2Z细胞代谢MTT的能力下降 ,显示明显的毒性反应。结论 :APBMV可能具有膜毒素的作用 ,通过降低CNE 2Z细胞膜电位 。 Aim: To observe the effect of APBMV on CNE 2Z cell membrane potential. Methods:The method of intracellular recording with glass microelectrodes was used to detect the membrane potential of CNE 2Z cells and MTT method to detect the cytotoxic effect of APBMV on CNE 2Z cells. Results: After CNE 2Z cells were treated with APBMV for 48 h, the negative potential of cell membrane was (13.00±3.58) mV,which was significantly lower than that of control (23.00±8.21) mV, (P < 0.01). Cell membrane was in depolarization state. The ability to metabolize MTT of CNE 2Z cells decreased. The cells showed distinct cytotoxicity. Conclusion:APBMV probably possesses the effects of the 'membrane toxin', which can interfere with the proliferation of tumor cells by decreasing the membrane potential.
