摘要
本实验采用抗原联接技术,以牛甲状腺球蛋白(BTG)为载体的前列腺素E_2(PGE_2)免疫原免疫C_(57)BL/6小鼠,1周后接种Lewis肺癌(LLC)。观察了抗-PGE_2抗体与内源性PGE_2作用后,对带瘤宿主免疫功能的影响。同时比较了前列腺素合成酶抑制剂消炎痛(INDO)的作用。结果表明,PGE_2抑制带瘤小鼠的免疫功能,抗-PGE_2和INDO能增加脾细胞对~3H-TdR的摄取、提高带瘤小鼠Th细胞数和NK细胞活性,并可明显降低实验组LLC组织中PGE_2含量。提示抗-PGE_2和INDO可阻遏或减轻PGE^2对带瘤小鼠免疫功能的抑制作用。
Antigen binding technique was employed with bovine thyroglobu-lin(BTG) as the carrier linked to prostaglandin E_2 (PGE_2) to im-munize C_(57) BL/6 mice, which were inoculated Lewis lung carcino-ma(LLC) cells one week later. We observed the effect of the anti-bodies against PGE_2 (anti-PGE_2), after interacting with the endo-genous PGE_2, on the immune function of the mice bearing LLC.In the meanwhile, the effect of a prostaglandin synthesis inhibitorindomethacin(INDO) was compared with. The results showed thatantiPGE_2 and INDO enhanced the ~3H-TdR incorporation of splenocy-tes, increased Th lymphocytes and NK cell activities. Radioimmuno-assay revealed a much lower content of PGE_2 in tumor tissues in theexperimental groups. Our research suggested that treatment of anti-PGE_2 and INDO could effectively prevent or alleviate PGE_2-mediatedimmune inhibition in tumor-bearing mice.
