摘要
目的在前期工作的基础上进一步将人源性抗HFRS病毒抗体轻链基因转染到鼠骨髓瘤细胞Sp2/0并进行初步表达。方法将含人源性抗HFRS病毒抗体轻链基因的真核表达载体VκEpSV-neo,用脂质体法转染鼠骨髓瘤细胞Sp2/0用RT-PCR法和免疫组化染色法检测转染及表达结果。结果RT-PCR和免疫组化染色结果表明人源性抗HFRS病毒抗体轻链基因不仅已成功地转染进Sp2/0细胞,并得到了良好的表达。结论在鼠骨髓瘤细胞Sp2/0中成功表达人源性抗HFRS病毒抗体轻链基因,对抗HFRS病毒抗体人源化的研究具有重要意义。
Aim To express the light chain gene of hum an anti -HFRS virus antibody in Sp2/0cells.Methods Eukaryotic expression vector VκEpSV -neo carrying light chain gene o f human anti -HFRS virus antibody was transfect into Sp2/0ce lls by lipofectin.RT -PCR and im-munohistochemical staining were used to detect the results of transfection and expression.Results It was showed that the light chain gen e of hu-man anti -HFRS virus antibody was successfully transfected into Sp2/0cell and expressed in it.Conclusion It is suggested that expressing the light chain gene of human anti -HFRS v irus antibody in eukaryotic cells is useful to humanize antibody.
