摘要 目的对比融合重组质粒CSpcDNA3.1单独注射,与SpcDNA3.1+CpcDNA3.1联合注射的免疫效果。方法大量提取质粒后免疫小鼠,剂量为每次150μg,按0、2和4wk,共免疫3次。免疫后第1、3、5和7wk采血,用ELISA方法检测抗HBs和抗HCV抗体;用LDH法检测免疫小鼠脾细胞CTL的杀伤功能。结果无论从抗HBs和抗HCV抗体出现的时间、阳转率和应答强度,还是CTL水平,CSpcDNA3.1融合重组质粒都优于CpcD-NA3.1+SpcDNA3.1联合基因的免疫。结论融合DNA疫苗,有望成为慢性病毒性肝炎预防和治疗的制剂。 Aim To compare the immunzation effect of the chimeric CSpcDNA3.1and the combination of SpcDNA3.1and CpcDNA3.1.Methods The mice were immunized three times with the chimeric and the combinated plasmids in 0,2and 4wk.T he dose was 150μg.The serum was collected in 1,3,5and 7wk..The anti -HBs and anti -HCV antibodies were detected by ELIS A and the cytotoxicty of spleno-cytes(CTL)from immunized mice was examined by L DH(lactate dehy-drogenase).Results Immumization of chimeric plasmid CSpcDNA3.1gained an advantage over that of comb inated CpcDNA3.1+SpcDNA3.1in the time to be found,detected positive rate and responsive intensity o f anti -HBs and anti -HCV antibodies.Conclusion Chimeric DNA vac-cine may become a promising agents of preventing and treating chronic viral hepatitis.
