摘要
目的 研究云芝子实体中多糖的化学结构。方法 应用柱色谱方法进行分离、纯化得一均一多糖组份B 1 3。通过完全水解、甲基化、一维和二维1 H和1 3 CNMR阐明B 1 3的化学结构。结果 经完全水解证实B 1 3主要由Glc单糖残基组成 ,用HPLC方法测得其分子量为 3 16× 10 5,通过对甲基化反应数据和1 H和1 3 CNMR分析发现B 1 3为由 β D 1,4 Glc和 β D 1,3 Glc残基构成主链的葡聚糖 ,同时在 β D 1,3,6 Glc及 β D 1,4,6 Glc残基处产生分枝。经1 H和1 3 CNMR分析表明B 1 3分子中存在 β D Glc 1→ 3 β D Glc β D Glc 1→ 4 β D Glc及 β D Glc 1→ 4 β D Glc β D Glc 1→ 4 β D Glc片段。结论 云芝子实体多糖B 1 3为由 β D 1,4 Glc,β D 1,3 Glc和 β D 1,6 Glc残基构成主链的葡聚糖。
AIM To study the structure of polysaccharides from Coriolus versicolor . METHODS By chromatography a homogeneous polysaccharide B-1-3 was obtained. The structure of B-1-3 was elucidated using the method of total acid hydrolysis, methylation, and 1D and 2D NMR. RESULTS B-1-3 was found to be mainly composed of Glc by total acid hydrolysis. Its molecular weight was estimated to be 3 16×10 5 by HPLC method. Methylation analysis and 1H and 13 CNMR indicated the main chain of B-1-3 consisted of β- D -1,4-Glc and β- D -1,3-Glc, and branch chains were situated, β- D -1,3,6-Glc and β- D -1,4,6-Glc. 1H and 13 CNMR also showed that β- D -Glc-1→3-β- D -Glc-β- D -Glc-1→4-β- D -Glc and β- D -Glc-1→4-β- D -Glc-β- D -Glc-1→4-β- D -Glc fragments existed in B-1-3. CONCLUSION B-1-3 is a glucan which main chain was composed of β- D -1,4-Glc and β- D -1,3-Glc β- D -1,6-Glc.
