大鼠急性全脑缺血再灌注细胞凋亡及白细胞介素1-β转化酶基因表达变化

摘要 目的:探讨急性全脑缺血再灌注后细胞凋亡及白细胞介素1-β转化酶(ICE)表达变化。方法:“四血管闭法”复制大鼠全脑缺血再灌注模型,应用流式细胞术(FCM)检测细胞凋亡,应用逆转录聚合聚链式反应(RT-PCR)技术检测ICEmRNA表达。结果:缺血30min再灌注,随缺血再灌注时间延长,DNA断裂百分率增高,再灌注24h达高峰,ICEmRNA表达呈增强趋势,再灌注6h达高峰,再灌注24h表达呈下降趋势,但仍表达较高水平。结论:ICE参与大鼠急性全脑缺血再灌注神经细胞凋亡的调控,其表达增强,促进神经细胞凋亡。 Objective:To study the effect of interleukin 1 beta converting enzyme (ICE) and apoptosis in rat with global cerebral ischemia and reperpfusion. Method: The global cerebral ischemia and reperfusion model was made by 4 vascular obstruction, using flow cytometry(FCM) to detect apoptosis and using reverse transcriptase-polymerase chaim reaction(RT-PCR) to detect ICE mRNA. Result: During reperfusion after 30 min ischemia with the passage of time, the rate of DNA fragmentation increases,and remained at high level after 24h reperfusion. The expresion of ICEmRNA increased gradually. The experession of reperfusion decreased after 6h reperfusion remained at high level and after 24h, but it remained at high level. Conclusion: ICE plays a vital role in regulating the neural apoptosis during reperfusion after ischemia. ICE level is high after cerebral ischemia and it promotes neural apoptosis.
