摘要
目的 建立测定神经中枢氨基酸类介质的方法。方法 邻苯二甲醛柱前衍生 ,ODS(C18)柱分离 ,pH6 0醋酸盐缓冲溶液与甲醇二元梯度洗脱 ,荧光检测。结果 仅在 9min内就分离测定了大鼠脑中 5种氨基酸介质的含量。在一定浓度范围内 ,氨基酸的峰面积与其浓度呈现良好的线性关系 ,相关系数在 0 .9978~ 0 .9999之间。
Objective To set up a method for determining the amino acid neurotransmitters. Methods o-phthaldialdelhyde/β-mercaptoethanol was used for the derivatization of amino acids.The separation of amino acid derivatives was accomplished on a column packed with 5 μm ODS. An acetate buffer pH 6.0 elution was adopted with a fluorescent detector (E x=330 nm,E m=456 nm). Results Five amino acids were separated and assessed merely in 9 min. Analysis of the results revealed a good linear relationship existing between the peak area and the concentration of amino acids. The correlation coefficients are in the range of 0.997?8~0.999?9. Conclusion The method is good enough for neuropharmacological research work.
