摘要
为了构建及表达抗人红细胞单链抗体 -汉滩病毒核衣壳截短抗原双功能蛋白 ,用于肾综合征出血热病人血清中抗HFRSV抗体的快速检测 ,我们将抗人红细胞的鼠单克隆抗体重、轻链可变区基因及汉滩病毒S基因片段N端部分基因克隆在同一个表达载体中 ,构建成分泌型双功能抗体表达载体pRBC -SN ,转化大肠杆菌XL -Blue,IPTG诱导表达。经ELISA测定 ,证明表达的双功能蛋白 ,具有抗人RBC和结合抗HFRSV抗体的双重功能。结果提示 ,表达的双功能蛋白具有双重活性 ,用于血清学快速检测 ,还需作进一步改构。
In order to construct and express the anti-RBC -truncated Hantaan virus nucleocapsid protein bifunctional molecule for rapid detection of serum anti-HFRSV antibody of patient's blood,the VL and VH genes of anti-RBC and the truncated S genome segment of Hantaan virus were cloned into the same expression vector to form a secreted bifunctional expression vector pRBC-SN.The bifunctional molecule was expressed in E.coli after transforming the XL-Blue and being induced by IPTG.The bifunctional molecule was proved by ELISA to be able to bind both RBC and anti-HFRSV antibody.The results show that the expression protein has double function,but it still needs modifications before it can used in rapid serum detection.
