摘要 目的 探讨激光间质内热疗诱导的效应灶的形态学变化和效应灶周围血脑屏障的改变。方法 利用正常鼠脑进行激光间质内热疗(ILTT),应用光学显微镜、电子显微镜等观察ILTT后效应灶形态学变化,伊文思蓝观察效应灶周围血脑屏障的改变。结果ILTT后效应灶急性期呈现显著的形态学变化:中央出血性假腔、凝固性坏死区、外周水肿;其后出现特征性的修复反应。效应灶周围血脑屏障暂时开放;细胞和亚细胞质膜的破坏在效应灶的形成过程中可能发挥重要作用。结论ILTT能够导致效应灶组织凝固性坏死和开放效应灶周围组织血脑屏障,这些结果有助于未来脑肿瘤激光间质内热疗的临床应用。 Objectives To explore morphologic changes of lesion interstitial laser thermotherapy induced and modification of the blood-brain barrier ( BBB) in the periphery of the lesion. Methods The lesion was induced in the normal rat brains by ILTT, morphologic changes of ILTT-induced lesion were determined by light microscopic and electron microscopic methods, triphenyltetrazolium chloride(TTC) stain, and modification of the blood-brain barrier in the periphery of the lesion was surveyed through Evens blue stain. Results The acute ILTT-induced lesions showed a distinct morphological zonal architecture: a central haemorrhagic pseudo cavity, coagulation zone and edematous zone. The tissue reaction followed the course known for coagulation necrosis and resulted in characteristic healing. The blood-brain barrier in the periphery of the lesion was temporarily opened. Membrane destruction on a cellular and subcellular level appears to be of major significance in the pathogenesis of the laser lesion. Conclusions ILTF could result in coagulation necrosis of the lesion and open blood-brain barrier in the periphery of the lesion, these results might be helpful in future clinical applications of ILTT for brain tumor.
