次声作用下大鼠脑皮层Ⅰ、Ⅱ组代谢性谷氨酸受体mRNA表达及意义

摘要 目的 探讨8Hz、130dB次声作用后Ⅰ、Ⅱ组代谢性谷氨酸受体(mGluR)mRNA在大鼠脑皮质的表达及意义。方法120只大鼠随机分为对照组及次声作用1、7、14次组,次声作用组采用自制的次声压力仓,用8Hz、130 dB的次声,每次作用 2h。 利用地高辛标记寡聚核苷酸探针原位杂交技术与计算机图像分析系统,在光镜与透射电镜下观察Ⅰ、Ⅱ组 mGluR mRNA在大鼠脑皮层阳性神经元的表达,以及病理与超微结构变化。结果 次声作用1次组,脑皮质内Ⅰ组mGluR的 mRNA阳性神经元数量增加;至 7次组增加至峰值(P<0.01);14次组基本恢复到正常。而Ⅱ组mGluR的mRNA表达与Ⅰ组mGluR变化趋势相反。光镜下.两组mGluR阳性神经元散在分布在脑皮质浅层,在胞浆和胞核内mRNA是均匀、颗粒状表达。结论 次声脑损伤后.I组 mGluR的 mRNA表达增强,而Ⅱ组 mGluR mRNA表达减弱,已知两组 mGluR对神经元分别具有损害和保护的不同作用,因此 Ⅰ、Ⅱ组 mGluR mRNA的变化参与了次声脑损伤作用。 Objective To study the alterations of metabolic glutamote receptor(mGluR) group Ⅰ,Ⅱ mRNA in brain cortex after infrasonic damage in rats, so as to investigate the function of mGluR group Ⅰ, Ⅱ in infrasonic damage. Methods l20 SD rats were randomly divided into four groups: control group, I-time, 7-time and l4-time infrasonic damage groups. Rats were exposed to 8 Hz l30 dB infrasound as designed, 2 h each time. mGluR group Ⅰ,Ⅱ mRNA was detected by digoxin-labeled oligonucleotide probe in situ hybridization method. The alterations of mGluR group Ⅰ, Ⅱ mRNA in cortex were observed under computer aided image analysis and light microscope. Result The expression levels of mGluR group I mRNA positive neurom increased in cortex after infrasonic damage in l-time group and was higher in 7 -time group (P < 0. 0l ). While the number of mGluR group I mRNA positive neurons in l4 time group reached norma1 level. On the contrary, the cxpression levels of mGluR group Ⅱ mRNA positive neurons decreased in cortex after infrasonic damage and was lower in l time group. mRNA positive neurons in both groups existed in superficial cortex. With light microscope observation, uniformly granulated mRNA was presented in the cytoplasm and nucleus. Conclusion Infrasound can induce an increase of group Ⅰ mRNA and an decrease of group Ⅱ mRNA expression in brain cortex.
