摘要
目的 探讨前列腺癌中P5 3蛋白和增殖细胞核抗原 (PCNA)表达的意义。②方法 用免疫组织化学技术 ,对 30例前列腺癌和 2 0例良性前列腺增生组织中P5 3蛋白和PCNA进行检测。③结果 6 3.3%前列腺癌中存在P5 3蛋白的异常表达 ,良性前列腺增生组织P5 3蛋白均表达阴性 ;前列腺癌PCNA增殖指数明显高于良性前列腺增生组织 (t′ =8.31,P <0 .0 0 1) ,P5 3蛋白及PCNA表达均与前列腺癌的组织学分级及病人预后有关 (P5 3:Hc=7.73,P <0 .0 5 ;z=2 .5 5 ,P <0 .0 5 .PCNA :F =36 .2 5 ,q =3.2 5~ 8.96 ,P <0 .0 1.t=5 .2 5 5 ,uc=2 .843,P <0 .0 0 1) ,前列腺癌P5 3蛋白阳性表达者PCNA增殖指数明显高于P5 3蛋白阴性表达者 (t=4.46 6 ,P <0 .0 0 1)。④结论 p5 3基因突变可能是促进前列腺癌发生发展和导致肿瘤细胞增殖异常活跃的重要因素 ;P5 3蛋白表达与PCNA增殖指数可作为鉴别前列腺良恶性肿瘤 ,以及判断前列腺癌的恶性程度和预测预后的综合性指标之一。
Objective\ To study the significance of P53 protein and proliferating cell nuclear antigen(PCNA) expressions in prostatic carcinoma. \ Methods\ Expressions of P53 protein and PCNA were detected in 30 cases of prostatic carcinoma and 20 cases of benign prostate hypertrophy(BPH) tissue by means of immunohistochemical method.\ Results\ 63.3% of prostatic carcinoma was positive for P53 protein expression, while 20 cases of BPH tissue were all negative; PCNA index was much higher in tumor tissue than that in BPH (t′=8.31,P <0.001). The expression of P53 protein and PCNA were related to tumor histological grading and prognosis(P53: H c=7.73,P<0.05; z=2.55,P <0.05. PCNA: F=36.25, q=3.25-8.96,P<0.01. t=5.255,u c=2.843,P <0.001). The PCNA index in positive cases expressing P53 protein was much higher than that in negative with the PCNA index in prostatic carcinoma( t=4.466,P <0.001). Conclusion\ P53 mutation might contribute to the development of prostatic carcinoma and enhance the proliferation rate of prostatic carcinoma cells;P53 protein expression and PCNA index could be used as some of the criteria in distinguishing malignant from benign neoplasms of the prostate and predicting prognosis of the disease.
