摘要
利用紫外差分光谱和可见光谱对产自嗜热放线菌M 10 3 3菌株的葡萄糖异构酶进行了研究 .结果表明 ,葡萄糖异构酶与其金属激活离子Co2 + 、Mg2 + 及抑制离子Ca2 + 存在键合作用 ,这种键合作用至少存在强键和弱键两种不同的结合方式 ,其结果使得葡萄糖异构酶分子结构发生了变化 .Co2 + 与酶分子有 8个强键结合位 ,6个弱键结合位 ,而Mg2 + 和Ca2 + 则分别有 3 2个和 2 6个强键结合位 .这 3种金属离子在酶分子上有一些相同的结合位 ,结合物的微环境中存在色氨酸或酪氨酸残基 .
Glucose isomerose from thermophilic actinomycetes M-1033 was studied by means of UV difference spectrum and visible spectrum.The results indicated that there is binding interaction between glucose isomerase and such activating cations as Co 2+ ,Mg 2+ and inhibiting cation Ca 2+ ,and by which structure of the engyme is changed.At least two different binding ways,tightly binding,and weakly binding,exist between the cations and the glucose isomerase.The enzyme has 8 tightly bound sites and 6 weakly bound sites to Co 2+ ,but 32 and 26 tightly bound sites to Mg 2+ and Ca 2+ .There are some same binding sites for the three cations with the glucose isomerase,and trp and tyr exist in the mini enviroment of the compounds.
