将活化长臂生物素(BCNHS)标记马铃薯卷叶病毒(Potato leaf-roll virus,PLRV)抗体,和辣根过氧化物酶,碱性磷酸酯酶标记链霉亲和素用于酶联免疫吸附试验(ELISA)中,成功地鉴定了PLRV。将ABC ELISA、BA ELISA和常规直接ELISA三者做了比较。三者对提纯的PLRV最低可测出浓度分别为1.25ng/ml,2.5ng/ml和20ng/ml,可测出感病马铃薯茎叶汁液中PLRV的最高稀释度分别为1:256、1:128和1:16。BA ELISA和ABC ELISA比常规直接ELISA的灵敏度分别提高8倍和16倍,而且操作简便,节省时间。
The biotin-avidin system (BAS) was successfully used in ELISA for the detection of potato leaf-roll virus ( PLRV ) . The antibody to PLRV was labelled with active space arm biotin (BCNHS) in this system. Two BAS ELISA designs were studied and compared with direct ELISA in which the antibody was labelled with alkaline phosphatase. When direct ELISA, BA ELISA and ABC ELISA were used,the lowest detectable concentrations of purified PLRV were 20ng/ml, 2.5ng/ml, and 1.25ng/ml respectively; and the highest detectable dilutions of extract of infected potato plants were 1:16, 1:128, and 1:256 respectively. BA ELISA and ABC ELISA were more sensitive than direct ELISA by 8 and 16 times respectively. They also offer other advantages over direct ELISA, notably in simplicity of antibody labeling and speed of development.
