摘要
鹰嘴豆和无壳葫芦籽黄化苗中苯丙氨酸银氨酶(PAL)经亲和层析纯化,聚丙烯酸胺凝胶电泳和SDS—FAGE鉴定均为一条蛋白带,用SDS-PAGE测得鹰嘴豆和无壳葫芦籽PAL亚基分子量分别为55000和74000。Sephadex G—200凝胶过滤法测得鹰嘴豆和无壳葫芦籽PAL分子量分别为205 000和280 000。表明这2科植物PAL均由分子量相同的4个亚基构成。
Gram chickpea and bottle gourd PAL have been isolated and purifiedby ammonium sulphate fractionation, column chromatographies on DEAE-cellulose-52 and L-phenylalanyl-Sepharose 4B. The purified preparationswere shown to be homogeneous by polyacrylamide gel electrophoresis inthe presence and absence of sodium dodecyl sulphate (SDS). The molecularweight of bottle gourd PAL subunit is 74000 dalton and that of gramchickpea PAL subunit is 55000dalton as assayed by SDS-polyacrylamidegel electrophoresis. The molecular weight of gram chickpea PAL and thatof bottle gourd PAL were found to be approximately 205000 dalton and280000 dalton by Sephadex G-200 gel filtration respectively. Thus, thesefindings showed that the native enzyme was composed of four subunits ofthe the same size.
