摘要
本文就痘苗病毒天坛株血凝素(VVHA)基因的核苷酸序列进行了分析,后者位于Hind Ⅲ A片段的Sal Ⅰ~Xba Ⅰ区段中,全长1391bp。自1至945bp有一长的ORF,编码34734道尔顿多肽,推导的多肽具有典型的穿膜蛋白的性质,成熟分子中有6个糖化位点(Asn-X-Thr/Ser)。VVHA分子中OH基氨基酸(Ser/Thr)含量很高,占28%,VVHA启动子具有痘苗病毒晚期启动子的特点,在启动子序列内部有一回文结构ACAAAAT/ATTTTGT,中间有5个核苷酸间隔。分子内部在核苷酸水平上(508~720)和氨基酸水平上(170~240)有一长的直接串联重复区,与WR株比较,在核苷酸水平上,结构基因有1.2%的变异,在氨基酸水平上有2.5%的变异。
The complete nucleotide sequence of the hemagglutinin ( HA ) gene of vaccinia virus Tian Tan strain in a 1 391 bp Sal Ⅰ-Xba Ⅰ subfragment of the Sal Ⅰ G fragment was determined by the Sanger dideoxy chain termination method. A single long ORF with 945bp ( 315aa)from 1 to 945bp was found. The deduced amino acid sequence reveals a structure typical for a trans-membrane glycoprotein. There are 5 stroqg N-glycosylation sites (Asn-X -Thr/Ser ) in the mature protein molecule.The HA molecule is rich in (?)mino acids with OH-group ( Thr/Ser ) ( 28% ) . The HA promoter sequence Is characteristic of the vaccinia virus late function. A palindromic se-quence ACAAAAT/ATTTTGT is located within its promoter region. Two direct tandem repeat sequences exist at nucleotide level from 508 to 720 and at amino acid level from 170 to 240.The variation between the Tian Tan and Western Reserve strains was detected at the nucleotide level with a rate of 1.2% and at the amino acid level with a rate of 2.5%.
