摘要
目的 探讨低剂量辐射 (LDR)诱导蛋白的产生条件、提取、检测、影响其生物活性的因素等 ,并进一步探讨其对淋巴细胞CD2 5分子表达的影响。方法 雄性昆明小鼠给予 5~ 10cGyLDR(γ射线 )全身照射后 2h ,取脾脏制成单细胞悬液 ,超声波破碎细胞、低温高速离心 ,上清液即为LDR诱导蛋白粗提液。以3H -TdR掺入法 ,观察LDR诱导蛋白对离体小鼠脾脏细胞转化的刺激作用以反映其生物活性。用间接免疫荧光—流式细胞仪测定方法 ,观察LDR诱导蛋白对人淋巴细胞CD2 5分子表达的影响。结果 经 5~ 15cGyLDR(γ射线 )作用后都可从小鼠脾脏细胞中提取到有生物活性的LDR诱导蛋白 ,并且在 10cGy剂量时LDR诱导蛋白活性最强 ,提取诱导蛋白的最佳时间在照后 2h左右。结论 LDR诱导蛋白能轻度增强静止的淋巴细胞CD2
Objective To find the substantial basis for effects of low dose radiation,on development,extraction,and the bidogical activity of the low-dose radiation-induced proteins,and the effects of LDR induced proteins on CD25 molecule expression of human lymphocytes.Methods 1.Healthy Kumning male mice exposed to radiation of 226 Ra r-rays at 5,10 and 15cGy respectively.The mice were killed 2 hours after exposure, the spleen cells were broken with ultrasonic energy and then ultracentrifugalized at low temperature(4℃).The LDR-induced proteins were obtained in the supernatant solution.Then the changes of CD25 molecule was measured by flow cytometry(FCM) with immunofluorescence technique,which was used to reflect the effect of LDR induced proteins on CD25 molecule expressionof human lymphocytes.Results LDR induced proteins were obtained from spleen cells in mice exposed to 5-15cGy whole body radiation.Condusion The expression of CD25 molecule of lymphocytes was increased significantly after use of LDR induced proteins. LDR induced proteins can enhance expresson of CD25 molecule of lymphocytes slightly.
