摘要
本实验观察了P4噬菌体在大肠杆菌C117中的增殖过程。该噬菌体在60℃几乎完全被灭活,对紫外线有抗性,对乙醇和EDTA溶液敏感。用电镜和琼脂糖凝胶电泳对其DNA进行分析提示,P4噬菌体中除含一般的双链开环和线性DNA外,在未完全成熟时它的头部衣壳中还有一种复杂的不均一打结DNA,这种特殊构型DNA的发现将为DNA拓扑异构酶Ⅱ的鉴定及酶活力测定奠定基础。
Bacteriophage P4 is a satellite virus, deponding on a helper genome of P2 phage for the completion of its lytic life cycle. The optimal growth of 'the phage, as well as its sensitivity to UV light and alcohol, EDTA, were determined. P4 DNA was extracted from mature phages or head , capsids. The majority of P4 DNA from mature phages appeared as normal linear and circular forms, whereas nearly all P4 DNA from head capsids appeared as highly knotted structure under the electron microscopy. Analysis of this knotted DNA by electrophoresis showed that knotted DNA molecules migrated in the gel as a wide band but exhibited the same molecular weight.
