特异寡聚核苷酸对猪瘟病毒在细胞中增殖抑制作用的研究 被引量:1

摘要 本实验探讨了寡聚核苷酸对CSFV复制的影响以及作为抗CSFV新型药物的可行性。实验结果表明针对CS FV 5’端非编码区NS3蛋白丝氨酸蛋白酶功能区的寡聚核苷酸对CSFV复制均有显著的抑制作用 ,而针对CSFV 3’端非编码区寡聚核苷酸仅有轻微抑制作用 ,5′端寡聚核苷酸具有最佳抑制作用 ,同时发现相应序列中正义聚核苷酸的作用要优于反义寡聚核苷酸 ;脂质体介导转染能显著提高寡聚核苷酸对CSFV复制的抑制作用。这些初步结果也提示 ,CSFV 5’端和 3’端非编码区对CSFV复制的重要性和作用有所不同。 The effect of oligodeoxynucleotides(ODN) on replication of CSFV(Shimen strain) was investigated in this paper.Phosphorthioate 01igodeoxynucl-eotides (PS-ODN) targeted to the region of the 3',5'-noncoding region(NCR) as well as PS-ODN targeted to NS3 gene region can markedly inhibited the replication of CSFV in PK-15 cells.Antiviral activity of PS-ODN was evaluated by fluorescent plaque formation assay.The results presented here indicated that ODN P5 complementary to 5'terminal conserved sequence and it's sense ODN P5S,as well as P8 complementar to NS3 region exhibited strongly antiviral activity,whereas the antiviral activity of P3 and it's enese PS-ODN was much lower.At the same time,antiviral activity of sense ODN is slightly higher than corresponding antisense ODN.The inhibition of ODN is in a dose-and sequence-dependant manner.Inhibition of CSFV's replication in PK-15 cells can be signfiicantly increased in the presense of lipofectin by either sense or antisense ODN.These data suggested both 5'-NCR and 3'-NCR may play important roles in CSFV replication.
