摘要
目的建立冰乙酸性大鼠胃溃疡模型并观察溃疡胃窦自愈过程中的组织学变化.方法将70只受试大鼠按每组7只随机分为10组,具体为第4天4个观察组(分别是0.01 m1、0.05 ml、0.10 m1冰乙酸组和0.05 ml生理盐水组)和第7、10、14、21、28天各1个观察组及正常对照组.各组大鼠依设计进行相应的处置,对实验大鼠进行外观表现观察、大体和光镜观察.结果注入0.01 ml冰乙酸的大鼠无明显溃疡,注入0.05 m1冰乙酸者出现典型胃窦溃疡,注入0.10 ml冰乙酸组发生胃穿孔.不同时间组大鼠的外观、胃窦大体和光镜观察呈现相应的溃疡表现和溃疡逐渐愈合的表现.结论大鼠胃窦前壁注入0.05 ml冰乙酸可制作典型的胃溃疡,溃疡胃窦粘膜于7~10 d后逐渐再生,14~28 d后逐步愈合.
Objective To establish an acetic acid-induced gastric ulcer model in rats and investigate the histological features of ulcerous antral mucosa. Methods A total of 70 Wistar rats were randomly divided into 10 groups (n=7) among which 4 groups, namely those with 0.01, 0.05 and 0.10 ml acetic acid and normal saline injections respectively, were observed on day 4, and group with 0.05 ml acetic acid injection, on day 7, 10, 14, 21 and 28 respectively and the left 1 group without any treatment served as control. An overview of the behaviors of the rats was conducted followed by gross and microscopic examinations of the rat antral mucosa. Results The rats with 0.01ml acetic acid treatment acquired no obvious gastric ulcer, while those with 0.05 ml acetic acid treatment suffered typical gastric ulcer, and the rats receiving 0.10 ml acetic acid injection even presented with gastric perforation. The microscopic observation showed 'a full spectrum of ulcerous changes in the antrum' in different groups. The ulcers eventually healed spontaneously over varied period of time. Conclusion Typical rat gastric ulcer model can be established by injecting 0.05 ml acetic acid into the rat antrum. The antral mucosa regenerates 7 to 10 d after the induction of the ulcers and spontaneous healing may occur eventually after14 to 28 d.
