摘要
从长白蝮蛇 ( Agkistrodon halys Ussurin)毒腺中抽提总 RNA,采用 RT-PCR扩增其类凝血酶基因 ,经全序列测定 ,获得 2个类凝血酶基因 ,ussurin和 ussurase,它们全长分别为 70 8和 699个核苷酸 ,即分别编码 2 36和2 33个氨基酸 ;根据同源性 ,推测它们的活性中心分别为His43 ,Asp88和 Ser1 82 与 His40 ,Asp85和 Ser1 79;二硫键分别为 Cys7-Cys1 4 1 ,Cys2 8-Cys44 ,Cys76 -Cys2 3 4,Cys1 2 0 -Cys1 88,Cys1 52 -Cys1 6 7和 Cys1 78-Cys2 0 3 ;与 Cys7-Cys1 3 8,Cys2 5-Cys41 ,Cys73 -Cys2 3 1 ,Cys1 1 7-Cys1 85,Cys1 4 9-Cys1 6 4 和 Cys1 75-Cys2 0 0 。该蛇毒类凝血酶 c
Total RNAs were extracted from the venom gland of the Agkistrodon halys Ussuriensis snake The thrombin like enzyme genes were amplified by RT PCR, cloned,and their nucleotide sequence have been determined The enzymes called ussurin and usurase cDNAs encode 708 and 699 nucleotides, namely 236 and 233 amino acids respectively Based on the homo1ogy, the catalytic residues and disulfide bridges of ussurin were deduced as follows: catalytic residues, His 43 , Asp 88 and Ser 182 ; and disulfide bridges, Cys 7 Cys 141 , Cys 28 Cys 44 , Cys 76 Cys 234 , Cys 120 Cys 188 , Cys 152 Cys 167 and Cys 178 Cys 203 ; and catalytic residues, His 40 , Asp 85 and Ser 179 ; and disulfide, Cys 7 Cys 138 , Cys 25 Cys 41 , Cys 73 Cys 231 , Cys 117 Cys 185 , Cys 149 Cys 164 and Cys 175 Cys 200 respectively
