摘要
用B细胞培养细胞系SB及Nalm-1作为异质性白血病细胞模型,与9倍数量正常骨髓细胞混合后,观察 CD9类抗体 DS21、CD10类抗体55对骨髓中 SB 及 Nalm-1细胞依赖补体的净化效果。用间接免疫荧光及溴乙锭双染法及集落形成实验均证明,联合使用 DS21及55抗体对靶细胞清除效果优于任何一个单一抗体,同时对骨髓中造血祖细胞无明显杀伤作用。本实验结果为临床联合应用 DS21及55抗体净化非 T-ALL 患者自体骨髓提供了依据。
Mixture of SB and Nalm-1 cells(1:1)was taken as leukemic heterogenous target cells and mixed again with 9 times of normal bone marrow cells.Then the purging effects of complement fixing McAbs 55,DS21 were observed.Using the indirect immunofluorescence and ethidium bromide(EB)dual staining as well as colony formation techniques,it was found that higher killing efficacy was obtained by using the combination of DS21 and 55 than using anyone of them.No obvious side effect on hematopoietic progenitor cells was found after the purging treatment of bone marrow.It is suggested that McAbs DS21 and 55 could be used clinically for purgation of residual leukemic cells from non-T-ALL pati- ents'autografts.
