摘要
本文应用放射性同位素分析方法,研究了[~3H]-T-2毒素给小鼠一次口服染毒后在体内的分布、蓄积和动力学过程。小鼠灌胃[~3H]-T-2毒素0.4mg/kg 后,在体内的吸收和分布都较快,血中及大部分组织中的放射活性在15min 到达高峰,然后呈双相下降。放射活性可以在胆囊和肝脏中浓集,大部分组织器官中的放射活性在染毒5d 后降至基线。毒素及其产物在脂肪中有一定的蓄积作用,脂肪球中的放射活性在染毒60d 后方完全消失。慢性染毒18个月的老年小鼠和正常对照老年小鼠,一次口服染毒[~3H]-T-2 毒素后,在毒素的体内处置上无显著性差异。[~3H]-T-2毒素的分布和毒物动力学过程也不存在年龄差异。
T-2 toxin is a 12,13-epoxytrichothecene mycotoxin produced by the Fusarium species. The distribution,accumulation and toxicokinetics of T-2 toxin were investigated in mice b radioisotope assay.The absorption and distribution of T-2 toxin were rapid after single do- sing 3~H-labeled T-2 toxin 0.4 mg/kg administered orally.The maximum radioactivity of plasma and most tissues were attained at 15min and the elimination of radioactivity was bi- phasic.Bile and liver contained a large amount of radioactivity.The patterns of distribution and excretion suggest that T-2 toxin and/or its metabolites are excreted into the intestine through the bile,and the liver is a major organ for excretion.Radioactivity of most tissues declined to the base line after 5 days and T-2 toxin or its metabolites hadsome specific ac- cumulation in fat.The disposition of T-2 toxin in the mice with chronic intoxication did not have singnificant change compared with the normal mice following single dose of the toxin. No age difference for distribution and toxicokinetics of T-2 toxin was found in mice.
