血管内皮细胞生长因子诱导大肠癌LOVO细胞粘附作用

摘要 目的 观察血管内皮生长因子 (VascularEndothelialGrowthFactor ,VEGF)对大肠癌LOVO细胞粘附作用。方法 采用3H-TdR掺入鼠尾胶粘附实验测定VEGF诱导大肠癌LOVO细胞同质性和异质性粘附作用。结果  1ng/mL、5ng/mLVEGF诱导LOVO细胞 6 0分钟、90分钟3H -TdR掺入实验 (dpm/min)分别为 1380 .0 3± 198.0 5 ,130 2 .2± 2 0 1.0 4和 2 6 41.6± 473 .2 4、2 797.5± 413 .2 4,10ng/mLVEGF诱导LOVO细胞 6 0分钟、90分钟、12 0分钟3H TdR掺入实验分别为 2 75 3 .0 3± 310 .31、1891.5 9± 2 2 3 .80、2 0 6 6 .79±187.97,显著低于对照组 6 0、90、12 0分钟的 1948.14± 2 16 .74、336 1± 175 .2 7、4173 .4± 373 .98,P <0 .0 5或 0 .0 1;5ng/mL、10ng/mLVEGF诱导LOVO细胞 6 0分钟鼠尾胶粘附实验OD值为 0 .2 5 0± 0 .0 18、0 .2 3± 0 .0 36 ,1ng/mL、5ng/mL、10ng/mLVEGF诱导LOVO细胞 90分钟、12 0分钟鼠尾胶粘附实验OD值分别为 0 .2 6 3± 0 .0 2 1、0 .373± 0 .0 83、0 .378± 0 .0 79和 0 .32 9± 0 .0 5 6、0 .389± 0 .0 95、0 .419± 0 .10 2 ,分别高于对照组的 0 .130± 0 .0 2 5、0 .14 3± 0 .0 36、0 .2 10± 0 .0 2 8,P <0 .0 5或 0 .0 1。 Aim To observe the effect on adhesion of large intestine cancer cell LOVO induced by Vascular Endothelial Growth Factor (VEGF).Methods We measured the function of homotypic adhesion and hetertypic adhesion in large intestine cancer cell LOVO With 3H-TdR infiltration and experiment of rats’tail colloid.Results After 60 and 90 minutes,the value(dpm/min) of 3H-TdR infiltration in LOVO cells induced by 1 ng/mL、5 ng/mL VEGF are 1380.03±198.05、1302.2±201.04 and 2641.6±473.24,2797.5±413.24,and that after 60,90 and 120 minutes by 10 ng/mL VEGF are 2753.03±310.31,1891.59±223.80 and 2066.79±187.97 respectively,which is dramatically lower than that in the control group, P <0.05 or 0.01.After 60 minutes,to OD value of the experiment of rats’ tail colloid in LOVO cells induced by 5 ng/ml and 10 ng/ml VEGF is 0.25±0.018,0.23±0.036.After 90 and 120 minutes,the OD values of the experiment of rats’tail colloid in LOVO cells induced by 1 ng/mL,5 ng/mL and 10 ng/mL VEGF are respectively 0.263±0.021, 0.373 ±0.083,0.378±0.079 and 0.329±0.056,0.389±0.095,0.419±0.102,which are remarkably higher than that in the control group, P <0.05 or 0.01.Conclusions VEGF can promote hetertypic adhesion and decrease homotypic adhesion of large intestine cancer cell LOVO.
