人视网膜组织中核因子-κB的免疫组织化学研究 被引量:1

Immunohistochemical study of NF-κB in the human retina

摘要 目的 :探讨正常和病理条件下人视网膜组织中核因子 (NF) κB的表达和活化。方法 :应用NF κB的特异性 p6 5单克隆抗体对 7只视网膜脱离和 2只继发性青光眼的人眼病理视网膜进行免疫组织化学观察 ,以 2只正常尸体眼视网膜作对照。结果 :9只病理眼视网膜均有细胞内NF κBp6 5阳性染色 ,NF κB活化 (胞核阳性染色 )主要见于内、外核层细胞 ,少数见于毛细血管内皮细胞和神经节细胞 ;2只对照眼视网膜细胞核内未见阳性染色。结论 :正常人视网膜组织中无NF κB活化 ,病理情况下视网膜中NF κB活性增强 ,提示NF Objective:To evaluate the expression and activation of nuclear factor (NF) κB in the human retina.Methods:Immunohistochemical detection was performed with a specific anti p65 mouse monoclonal antibody in the retina of seven eyes with retinal detachment and two eyes with chronic secondary glaucoma.Two retinas from cadaveric healthy eyes were assayed as normal control.Results:NF κB was expressed and activated in the cytoplasm and nucleus in the retinas of all nine pathologic eyes.Activation of NF κB occurred preferentially in the inner and outer nuclear layers,but less in the capillary endothelium cells in the detached retinas and the ganglion cells in the retinas of glaucoma eyes.No positive nuclear staining of NF κB was observed in the retinas of control eyes.Conclusions:NF κB is not activated in the normal human retina,but activated in the retinas of eyes with retinal detachment or glaucoma.NF κB may play a role in the pathogenesis of human retinal disease.
