摘要
目的 :观察鼻息肉中 IL- 5 m RNA的表达水平 ,并研究其与嗜酸性粒细胞浸润和活化状态的相关性 ,以期加深对鼻息肉发病机制的认识。方法 :采用逆转录 -多聚酶链反应 (RT- PCR)结合半定量分析方法 ,测定 16例鼻息肉中 IL -5 m RNA的表达水平。同时分别采用 chromotrope特染和免疫组化 (抗 EG2 )法 ,标记组织中的嗜酸性粒细胞和活化嗜酸性粒细胞。最后进行相关性分析。结果 :116例鼻息肉组织中均有 IL- 5 m RNA表达 ,而 10例中鼻甲组织中只有 1例显示 IL- 5 m RNA表达。 2变应性患者 (n=5 )和非变应性患者 (n=11) ,IL- 5 / β- actin光密度比值无显著性差异 (P >0 .0 5 )。IL- 5 / β- actin光密度比值与 chromotrope2 R阳性细胞密度相关 (y=- 30 .892 +10 7.5 30 x,r=0 .5 10 ,P<0 .0 5 ) ,与 EG2 阳性细胞密度的相关性不显著 (r=0 .376 ,P >0 .0 5 )。结论 :IL - 5 m RNA在鼻息肉组织中稳定表达 ,表达水平与嗜酸性粒细胞的浸润程度密切相关 ,说明组织中 IL - 5的基因表达状况与嗜酸性粒细胞浸润程度之间存在密切联系。
Objective:To assess the correlation between the expression of IL 5 mRNA and the infiltration and activation of eosinophils in nasal polyps.Methods:Nasal polyps from sixteen patients were investigated .Reverse transcription PCR(RT PCR)was used to investigate IL 5 mRNA expression.And the samples were stained with Chromotrope 2R histochemical and ABC immunohistochemical methods.Results:The bands of IL 5 mRNA were observed in all nasal polyp specimens,but only one of ten middle turbinate specimens.There was no difference in mean IL 5/β actin density ratio between allergic and nonallergic patients. And we found the mean IL 5/β actin density ratio correlated with the density of total eosinophils(y=-30 892+107 530x,r=0 510,P<0 05),but not that of activated eosinophils (r=0 376,P>0 05).Conclusion:The correlation was reported between IL 5 mRNA expression and numbers of chromotrope 2R positive cells ,which supported a link between IL 5 and eosinophils in nasal polyposis in vivo.
