摘要
目的 建立了高效液相 质谱联用法测定人血浆中单硝酸异山梨酯 (IS 5 MN)的浓度。方法 采用WatersC1815 0mm×2 .1mm色谱柱 ,以甲醇 氨水溶液 (0 .0 3mol·L-1) =2 0∶80 (pH =7.8)为流动相 ,流速为 0 .2 5mL·min-1,检测质荷比 (m/z)为190 ,血浆样品用醋酸乙酯直接提取 ,进样量为 2 0 μL。 结果 线性范围为 10~ 6 40 μg·L-1,最低检测浓度 7μg·L-1,日内、日间的RSD皆小于 8%。
OBJECTIVE: To establish a HPLC-MS method for the determination of IS-5-MN in human plasma. METHODS: IS-5-MN was separated on a Waters C18 column with the mobile phase consisted of methanol-(0.03 mol·L-1) ammonia solution. The flow rate was at 0.25 mL·min-1 .The ion mass spectral (m/z) was 190. The plasma samples were extracted by ethyl acetate. RESULTS: The linear range was 10 ∼ 640 μg·L-1 and the determintation limit was 7 μg·L-1. The extraction recovery of IS-5-MN was more than 80%. The within-day RSD and inter-day RSD were less than 8%. CONCLUSION: This method was found to be sensitive, fast, and accurate. Therefore, it can be used for the pharmacokinetic studies of IS-5-MN in human plasma.
