摘要
目的:通过建立大鼠脂肪肝模型,探讨脂肪肝在热缺血再灌注(I/R)急性损伤过程中血清丙二醛(MDA),丙氨酸转氨酶(ALT),内皮素-1(ET-1),肿瘤坏死因子-α(TNF-α)水平的变化。方法:首先复制脂肪肝模型,观察脂肪肝组和正常肝组门脉压的关系,以及在缺血前、缺血15分再灌注60分、再灌注24小时,肝组织中MDA、血清ET-1、TNF-α、ALT水平的变化。结果:脂肪肝组门脉压高于正常肝组。缺血前后脂肪肝组肝组织中MDA含量、血清ALT水平均高于正常肝组。血清ET-1、TNF-α水平在脂肪肝和正常肝组之间无显著性差异,但急性损伤后呈升高趋势。结论:肝脂肪变性引起肝窦狭窄和不规则,门脉压增高。含大量脂肪的肝细胞对热I/R损伤敏感性增高。
To investigate the changes of MDA, ALT, ET-1 and TNF-a in fatty liver rats with acute liver warm isch-emia-reperfusion (I/R) injury. Methods: First of all, fatty liver models of rat were produced. The relationship of portal vein pressure between fatty and normal liver groups and the changes of MDA, ALT, ET-1 and TNF-a before and 15 minutes after ischemia and during sixty minutes and twenty-four hours of reperfusions were observed. Results: The portal vein pressure in the fatty liver group was higher than that in the normal liver group. Before and after ischemia, the levels of MDA and ALT in fatty liver group was higher than that in normal liver group. There was no difference of levels of serum ET-1 and TNF-a between the two groups, but these levels increased after acute injury. Conclusion: Liver steatosis can result in narrow and irregular liver sinus and increase the portal vein pressure. The liver cells containing many fatty droplets are sensitive to acute liver warm I/R injury.
