摘要
目的 :探讨黄芪对脑缺血后脑细胞凋亡相关基因表达的影响。方法 :用结扎双侧颈内动脉的方法复制脑缺血模型 ,免疫组化和流式细胞仪分别检测bcl 2、p53免疫阳性细胞和凋亡细胞。结果 :结扎双侧颈内动脉后 ,脑局部血流量下降 ,脑皮质促凋亡基因表达明显增强 ,但未出现凋亡群体细胞。通过腹腔注射黄芪注射液后可抑制 p53表达、增强bcl 2表达。结论 :黄芪可抑制缺血区促凋亡基因的表达 ,增强抑凋亡基因的表达。
Objective:To investigate the effects of Astragalus on expression of apoptosis related genes after cerebral ischemia.Method:Cerebral ischemia model was made by ligation of both internal carotid artery.Immunohistochemical method and flow cytometry were used to observe bcl-2 and p53 positive immunocytes and apoptotic cells.Result:Cerebral blood flow was decreased,the gene controlling apoptosis in cerebral cortex was upregulated significantly after ligation of both internal carotid artery.But there was no existence of apoptotic cell group.Expression of p53 was inhibited,and that of bcl-2 was enhanced after astragalus was administrated by intraperitoneal injection.Conclusion:Astragalus could inhibit the expression of proapoptotic genes and enhance that of inhibiting-apototic genes in ischemia area after cerebral ischemia.
