通过根癌农杆菌 (Agrobacteriumtumefaciens)介导 ,利用特异表达的phaseolin基因的启动子与人类野生型抗癌基因p5 3构建嵌合基因 ,对‘美味樱桃’番茄进行遗传转化 ,获得转化番茄再生植株。通过组织化学法、PCR扩增及Southern杂交检测 ,证明人类野生型p5
The strain GV3101 of Agrobacterium tumerfaciens harboring a plasmid carrying a human wild type tumor suppressor gene p 53 driven by the promoter of phaseolin was used to transform cv.Meiwei Cherry of tomato.Leaves of transgenic tomato plants were used for assaying the existence of foreign gene.Results of histochemical assay,PCR amplification and Southern blotting showed that the human wild type p 53 gene was integrated into the genome of transgenic tomato plants.
