摘要
本实验用 4个酶、 4个探针组成 16个酶 /探针组合对玉米 N、T、C、S、WBMs、 80 1CMS等细胞质进行了线粒体 DNA( mt DNA) RFL P分析。一方面对玉米细胞质 mt DNA RFL P分类方法进行研究 ,证明只要酶 /探针技术体系合适 ,可以通过该方法对细胞质进行快速准确地分类 ;提出探针的选择是主要的 ,酶次之 ;认为 Pst /B30、H ind /p Bcm H3、Bam H /p HJ2 - 7- 1三个酶 /探针可以作为 mt DNARFLP分类技术体系。另一方面 ,用 mt DNA RFL P分类法把新发现的 WBMs不育胞质划分为 S组 ,80 1CMS划分为
RFLP analysis of mtDNA was carried out with 16(4×4) combinations of enzyme/probs to study the N T, C, S cytoplasmics of maize, as well as the uncertain cytoplasmics of a male sterils lines, WBMs and 801 Ms. The results indicated: (1) RFLP analysis of mtDNA could classify the cytoplasm of maize quickly and correctly if appropriate enzyme/probe. Combination(s) was(were) used. (2) Option of probes was of primary importance with that of enzymes ranking the second . (3) Three enzyme/probe combinations, i. e., Pst Ⅰ/B30, Hin dⅢ/pBcmH 3 and Bam HⅠ/pHJ2 7 1, were suggested as better technical systems for the classification of the cytoplasmics of maize with RFLP mtDNA analysis. (4) The male Sterile Cytoplasmics of WBMs and 801 Ms were classified as S and C cytoplasms respectively according to results of RFLP mtDNA analysis.
