摘要
目的 :探讨骨肉瘤特异性细胞毒T淋巴细胞 (OSS CTL)对人骨肉瘤细胞系HOS 860 3细胞凋亡的诱导作用 ,以揭示其抗肿瘤机制。方法 :OSS CTL与HOS 860 3细胞按不同效靶比共同孵育后 ,通过电镜观察HOS 860 3细胞的超微结构变化 ,并用流式细胞仪定量检测OSS CTL与HOS 860 3细胞凋亡发生状况。结果 :HOS 860 3细胞被OSS CTL攻击后浓缩、深染 ,致密的染色质沿核膜下凝聚 ,并有凋亡小体形成。效靶比为 1∶1、5 0∶1、10 0∶1时 ,HOS 860 3细胞凋亡发生率分别为 0 .6%、12 .4%及 2 1.6%。结论 :OSS CTL可通过诱导人骨肉瘤细胞系HOS 860 3发生细胞凋亡起到抗肿瘤作用 。
Objective:To discuss the OSS CTL induced apoptosis of HOS 8603 cells in order to reveal its antiumor mechanism.Method:OSS CTL and HOS 8603 cells were incubated together according different attack target cell ratio.Apoptosis of target cells were defected quantitatively with flowcytometry,the characteristic changes of morphology of HOS 8603 cell apoptosis were observed under electron microscope.Result:After attacked by OSS CTL,HOS 8603 cells showed the following changes,such as shrinkage,nuclear condensation and pyknosis.As attack target cells were 1∶1,50∶1,100∶1,the percentage of target cell were 0.6%,12.4% and 21.6%,respectively.Conclusion:OSS CTL had a high antitumor activity by inducing apoptosis of HOS 8603 cells.The amount of the apoptosis target cells obviously increased with the high attack target cell ratio.
