摘要
目的:观察小鼠胚胎脊髓(spinal cord,SC)和脊神经节(spinal ganglion,SG)发生中的细胞凋亡。方法:以体视学方法观察E12d~E15.5d ICR胎鼠H.E染色石蜡连续切片中臂丛段、胸段、腰从段的SC和SG的凋亡细胞密度(Density of apoptotic cell,DAC)。部分切片用TUNEL法标记凋亡细胞作对照。结果:胎鼠SC和SG中均见核碎裂的凋亡细胞。SC中的凋亡细胞主要分布在基板,偶见于翼板。统计结果显示,三段SC中的DAC峰值均在E14 d,P<0.01;臂丛段的DAC高于后两段,除E15d外,P均<0.05。胸段和腰从段SG中的DAC峰值均在E15d,P<0.01;臂丛段的DAC峰值不明显。结论:E12d~E15.5d胎鼠SC和SG中,凋亡细胞主要见于脊髓的基板和脊神经节。SC的DAC峰值较SG早一天。DAC的峰值可能与其组织分化有关。
Objective:To observe the apoptosis of developi ng spinal cord and spinal ganglion in the mouse embryo. Methods:The density of apoptotic cell(DAC) of the brachial plexus,thoracic and lumbar plexus of spinal cord (SC)and spinal ganglion (SG)in the serial sections of fetal ICR mice aged 12~15.5 days was measured with the stereological method.The apo ptosis was also showed with TUNEL method in some sections for contrast.Re sults:The apoptotic cells with karyorrhexis can be seen in SC and SG of fetal mice.The apoptotic cells of SC are mainly located in the basal plate,but rarely in the alar plate.The statistical results showed that the peak values of DAC in the three sections of SC all appeared on day 14,P<0.01; the peak value of DAC in the brachial plexus of SC was higher than the other two sections,P<0.0 5,except day 15.The peak values of DAC in the thoracic and lumbar plexus of SG all appeared on day 15,P<0.01;the DAC of brachial plexus was not clear.Conclusion :The apoptotic cells in SC and SG of fetal mice aged 12~15.5 days were mainly found in the basal plate of SC and the SG.The pea k values of DAC in SC were one day earlier than in SG.The peak values of DAC in the three sections of SC and SG may be related with their tissue-differeniation.
