摘要
安祖花嫩茎切段离体培养的结果表明 ,基本培养基都以改良的MS为最好 ;初代培养试验的四种激素浓度配比 ,均能诱导切段脱分化形成愈伤组织 ,诱导率最高的组合为改良MS +BA 1.0mg·L 1,诱导率达 88.17% ,培养 3 0d ,愈伤组织块的直径超过 1cm。愈伤组织转接于改良MS +BA 0 .8mg·L 1+NAA 0 .0 5mg·L 1培养基上 ,丛生芽的分化率在 80 %左右 ,丛芽月增殖系数为 2~ 3。将 1cm以上的小芽切下转接于改良MS +IBA 0~ 0 .1mg·L 1或NAA 0 .1mg·L 1促生根培养基上 ,6~ 7d发根形成完整植株。移栽基质以蛭石、椰糠、碎花泥为好 ,成活率在 85%以上。
The young shoot cutting culture and propagation in vitro of Anthurium andraeanum Lind were carried out. The results showed that the optimal mediums at various culture stages were always different improved MS mediums. Four kinds of improved MS mediums in initial culture all induced well callus formation. Among them, the improved MS+BA 1.0mg·L 1 led to the highest callus inducing rate(88.17%). After 30 days culture on this medium, the calli were all larger than 1 cm. When the calli were then transferred to the mediums of improved MS+BA 0.8 mg·L 1 +NAA 0.05 mg·L 1 , the bud differentiation rates were 80% or so. The monthly propagation coefficients of buds were 2~3 in the subculture of cut buds. The improved MS+IBA 0~0.1mg·L 1 or NAA 0.1 mg·L 1 had the best effect on inducing root formation. The plantlets formed could be transplanted easily, the survival rate was more than 85%.
